How to Reconstitute Peptides
A complete step-by-step protocol for reconstituting lyophilized peptide compounds with bacteriostatic water for laboratory research use.
What Is Reconstitution?
Lyophilized (freeze-dried) peptides are supplied as a dry powder to maximize stability during shipping and storage. Before use in a research setting, the peptide must be reconstituted — dissolved into a sterile solvent to create a usable solution. The most common solvent used is bacteriostatic water, which contains 0.9% benzyl alcohol to prevent microbial growth and extend the shelf life of the reconstituted solution.
What You Need
- Lyophilized peptide vial (e.g., BPC-157, Tesamorelin)
- Bacteriostatic water for injection
- Graduated research syringe (0.3 mL, 0.5 mL, or 1.0 mL)
- Alcohol swabs (70% isopropyl)
- Clean, sterile workspace
How Much Bacteriostatic Water to Use
The volume of bacteriostatic water added determines the final concentration of the solution. Use the Peptide Calculator to calculate the exact volume for your vial size and research aliquot target. A common starting point for laboratory preparations:
| Vial Size | BAC Water Added | Final Concentration | Per 0.1 mL Draw Volume |
|---|---|---|---|
| 5 mg | 1 mL | 5,000 mcg/mL | 500 mcg |
| 5 mg | 2 mL | 2,500 mcg/mL | 250 mcg |
| 10 mg | 1 mL | 10,000 mcg/mL | 1,000 mcg |
| 10 mg | 2 mL | 5,000 mcg/mL | 500 mcg |
| 10 mg | 4 mL | 2,500 mcg/mL | 250 mcg |
Step-by-Step Reconstitution Protocol
Prepare a Clean Workspace
Work in a clean, low-traffic area. Wipe down the surface with 70% isopropyl alcohol and allow it to dry completely before beginning.
Swab All Vial Tops
Wipe the rubber stopper of the bacteriostatic water vial and the peptide vial with a fresh alcohol swab. Allow both to air dry for 30 seconds before piercing.
Draw the Bacteriostatic Water
Using a clean syringe, draw the desired volume of bacteriostatic water from the BAC water vial. Use the Peptide Calculator to confirm the correct volume for your target concentration.
Introduce the Solvent Along the Vial Wall
Insert the needle into the peptide vial at an angle so the bacteriostatic water runs slowly down the inside glass wall — do not direct the flow onto the powder. This prevents peptide degradation from mechanical impact.
Swirl Gently — Do Not Shake
Gently swirl the vial in a slow circular motion until the powder is fully dissolved. Do not shake or vortex — agitation can denature the peptide and reduce potency.
Inspect the Solution
The reconstituted solution should be clear and free of particles. A slightly colored or cloudy solution may indicate contamination or incomplete dissolution — do not use if cloudiness persists after swirling.
Label and Refrigerate
Label the vial with the compound name, concentration, date reconstituted, and initials. Store refrigerated at 2–8°C. See the Peptide Storage Guide for detailed requirements by compound.
Never use regular sterile water for reconstitution — it does not contain a preservative and will allow microbial growth. Always use bacteriostatic water containing 0.9% benzyl alcohol for multi-dose vials.
Post-Reconstitution Storage
Once reconstituted, peptides should be stored refrigerated at 2–8°C and used within 28–60 days depending on the compound. Do not freeze reconstituted solutions. For complete temperature, light, and duration requirements, see the Peptide Storage Guide.
